The Rat Genome Database ( RGD ) is a database of rat genomics , genetics, physiology and functional data, as well as data for comparative genomics between rat, human and mouse. RGD is responsible for attaching biological information to the rat genome via structured vocabulary, or ontology , annotations assigned to genes and quantitative trait loci (QTL), and for consolidating rat strain data and making it available to the research community. They are also developing a suite of tools for mining and analyzing genomic, physiologic and functional data for the rat, and comparative data for rat, mouse, human, and five other species.

91-477: RGD began as a collaborative effort between research institutions involved in rat genetic and genomic research. Its goal, as stated in the National Institutes of Health’s Request for Grant Application: HL-99-013, is the establishment of a Rat Genome Database to collect, consolidate, and integrate data generated from ongoing rat genetic and genomic research efforts and make this data widely available to

182-750: A Special Cancer Investigations laboratory at Harvard Medical School . This marked the beginning of a partnership with universities. In 1930, the Hygienic Laboratory was re-designated as the National Institute of Health by the Ransdell Act , and was given $ 750,000 to construct two NIH buildings at the Old Naval Observatory campus. In 1937, NIH absorbed the rest of the Division of Scientific Research, of which it

273-406: A biological cell is adenosine triphosphate (ATP), which stores its energy in the phosphoanhydride bonds . The energy is utilized to conduct biosynthesis, facilitate movement, and regulate active transport inside of the cell. Examples of amphibolic pathways are the citric acid cycle and the glyoxylate cycle. These sets of chemical reactions contain both energy producing and utilizing pathways. To

364-399: A catabolic pathway provides the energy required to conduct the biosynthesis of an anabolic pathway. In addition to the two distinct metabolic pathways is the amphibolic pathway, which can be either catabolic or anabolic based on the need for or the availability of energy. Pathways are required for the maintenance of homeostasis within an organism and the flux of metabolites through

455-471: A certain percentage (2.8% in 2014) must be granted to small businesses under the SBIR/STTR program. As of 2011 , the extramural funding consisted of about 50,000 grants to more than 325,000 researchers at more than 3000 institutions. By 2018 , this rate of granting remained reasonably steady, at 47,000 grants to 2,700 organizations. In FY 2010 , NIH spent US$ 10.7bn (not including temporary funding from

546-439: A chemical bond, whereas a non-covalent modification (also known as allosteric regulation) is the binding of the regulator to the enzyme via hydrogen bonds , electrostatic interactions, and Van der Waals forces . The rate of turnover in a metabolic pathway, also known as the metabolic flux , is regulated based on the stoichiometric reaction model, the utilization rate of metabolites, and the translocation pace of molecules across

637-548: A clinical trial consisting of 185 adult patients with cholangiocarcinoma and an IDH-1 mutation, there was a statistically significant improvement (p<0.0001; HR: 0.37) in patients randomized to ivosidenib. Still, some of the adverse side effects in these patients included fatigue, nausea, diarrhea, decreased appetite, ascites, and anemia. In a clinical trial consisting of 199 adult patients with AML and an IDH2 mutation, 23% of patients experienced complete response (CR) or complete response with partial hematologic recovery (CRh) lasting

728-508: A clinical trial, except for the most critically ill, as determined by the NIH Director. Over the last century, the responsibility to allocate funding has shifted from the OD and Advisory Committee to the individual ICs and Congress increasingly set apart funding for particular causes. In the 1970s, Congress began to earmark funds specifically for cancer research, and in the 1980s there was

819-796: A coupled reaction is the phosphorylation of fructose-6-phosphate to form the intermediate fructose-1,6-bisphosphate by the enzyme phosphofructokinase accompanied by the hydrolysis of ATP in the pathway of glycolysis . The resulting chemical reaction within the metabolic pathway is highly thermodynamically favorable and, as a result, irreversible in the cell. Fructose − 6 − Phosphate + ATP ⟶ Fructose − 1 , 6 − Bisphosphate + ADP {\displaystyle {\ce {Fructose-6-Phosphate + ATP -> Fructose-1,6-Bisphosphate + ADP}}} A core set of energy-producing catabolic pathways occur within all living organisms in some form. These pathways transfer

And fifth, construct and support the scientific infrastructure (e.g., well-equipped laboratories and safe research facilities) necessary to conduct research. Advisory committee members advise

1001-495: A median of 8.2 months while on enasidenib. Of the 157 patients who required transfusion at the beginning of the trial, 34% no longer required transfusions during the 56-day time period on enasidenib. Of the 42% of patients who did not require transfusions at the beginning of the trial, 76% still did not require a transfusion by the end of the trial. Side effects of enasidenib included nausea, diarrhea, elevated bilirubin and, most notably, differentiation syndrome. Glutaminase (GLS),

Additional educational activities include the production of tutorial videos, both outlining how to use RGD tools and data, and on more general topics such as biomedical ontologies and biological (i.e. gene, QTL and strain) nomenclature. These videos can be viewed on several online video hosting sites including YouTube . RGD

1183-420: A pathway is regulated depending on the needs of the cell and the availability of the substrate. The end product of a pathway may be used immediately, initiate another metabolic pathway or be stored for later use. The metabolism of a cell consists of an elaborate network of interconnected pathways that enable the synthesis and breakdown of molecules (anabolism and catabolism). Each metabolic pathway consists of

1274-601: A rate similar to inflation since the 1970s, research funding for the NIH nearly tripled through the 1990s and early 2000s, but has remained relatively stagnant since then. By the 1990s, the NIH committee focus had shifted to DNA research and launched the Human Genome Project . The NIH Office of the Director is the central office responsible for setting policy for NIH, and for planning, managing, and coordinating

1365-477: A reaction. For example, one pathway may be responsible for the synthesis of a particular amino acid, but the breakdown of that amino acid may occur via a separate and distinct pathway. One example of an exception to this "rule" is the metabolism of glucose . Glycolysis results in the breakdown of glucose, but several reactions in the glycolysis pathway are reversible and participate in the re-synthesis of glucose ( gluconeogenesis ). A catabolic pathway

1456-400: A series of audit reports revealing that throughout the fiscal years 2000–2010, institutes under the aegis of the NIH did not comply with the time and amount requirements specified in appropriations statutes, in awarding federal contracts to commercial partners, committing the federal government to tens of millions of dollars of expenditure ahead of appropriation of funds from Congress. The NIH

1547-415: A series of biochemical reactions that are connected by their intermediates: the products of one reaction are the substrates for subsequent reactions, and so on. Metabolic pathways are often considered to flow in one direction. Although all chemical reactions are technically reversible, conditions in the cell are often such that it is thermodynamically more favorable for flux to proceed in one direction of

1638-523: A significant amount allocated for AIDS/HIV research. Funding for the NIH has often been a source of contention in Congress, serving as a proxy for the political currents of the time. During the 1980s, President Reagan repeatedly tried to cut funding for research, only to see Congress partly restore funding. The political contention over NIH funding slowed the nation's response to the AIDS epidemic; while AIDS

1729-436: Is a series of reactions that bring about a net release of energy in the form of a high energy phosphate bond formed with the energy carriers adenosine diphosphate (ADP) and guanosine diphosphate (GDP) to produce adenosine triphosphate (ATP) and guanosine triphosphate (GTP), respectively. The net reaction is, therefore, thermodynamically favorable, for it results in a lower free energy for the final products. A catabolic pathway

1820-402: Is an exergonic system that produces chemical energy in the form of ATP, GTP, NADH, NADPH, FADH2, etc. from energy containing sources such as carbohydrates, fats, and proteins. The end products are often carbon dioxide, water, and ammonia. Coupled with an endergonic reaction of anabolism, the cell can synthesize new macromolecules using the original precursors of the anabolic pathway. An example of

1911-560: Is available at the bottom of the individual disease portals. As of May 2021, RGD has fifteen disease portals: Disease portals consolidate the data in RGD for a specific disease category and present it in a single group of pages. Genes, QTLs and strains annotated to any disease in the category are listed, with genome-wide views of their locations in rat, human and mouse (see "Genome Viewer" in Genome tools developed at RGD ). 2002-613: Is available to the general public whereas the individual-level data is accessible to researchers. According to the City Journal NIH denies access to such attributes as intelligence, education and health on the grounds that studying their genetic basis would be stigmatizing. The NIH partnered with Moderna in 2020 during the COVID-19 pandemic to develop a vaccine. The final phase of testing began on July 27 with up to 30,000 volunteers assigned to one of two groups—one receiving

2184-443: Is determined by the scientist. The total number of applicants has increased substantially, from about 60,000 investigators who had applied during the period from 1999 to 2003 to slightly less than 90,000 in who had applied during the period from 2011 to 2015. Due to this, the "cumulative investigator rate", that is, the likelihood that unique investigators are funded over a 5-year window, has declined from 43% to 31%. R01 grants are

2275-667: Is funded by grant R01HL64541 from the National Heart, Lung, and Blood Institute ( NHLBI ) on behalf of the National Institutes of Health (NIH). The principal investigator of the grant is Anne E. Kwitek, who was appointed to this leadership position from Mary E. Shimoyama, in March 2020. Melinda R Dwinell is Co-Investigator. The new genome rat assembly, mRatBN7.2, was generated by the Darwin Tree of Life Project at

2366-445: Is non-spontaneous. An anabolic pathway is a biosynthetic pathway, meaning that it combines smaller molecules to form larger and more complex ones. An example is the reversed pathway of glycolysis, otherwise known as gluconeogenesis , which occurs in the liver and sometimes in the kidney to maintain proper glucose concentration in the blood and supply the brain and muscle tissues with adequate amount of glucose. Although gluconeogenesis

2912-427: Is the only GLS inhibitor currently undergoing clinical studies for FDA-approval. Many metabolic pathways are of commercial interest. For instance, the production of many antibiotics or other drugs requires complex pathways. The pathways to produce such compounds can be transplanted into microbes or other more suitable organism for production purposes. For example, the world's supply of the anti-cancer drug vinblastine

3003-403: Is unfavorable in a cell due to a positive Gibbs free energy (+Δ G ). Thus, an input of chemical energy through a coupling with an exergonic reaction is necessary. The coupled reaction of the catabolic pathway affects the thermodynamics of the reaction by lowering the overall activation energy of an anabolic pathway and allowing the reaction to take place. Otherwise, an endergonic reaction

3185-509: The Consolidated Appropriations Act, 2022 . Modeled after DARPA , HSARPA , IARPA , and ARPA-E , it is intended to pursue unconventional research projects through methods not typically used by federal agencies or private sector companies. Secretary Xavier Becerra delegated ARPA-H to the NIH on May 24, 2022. It received $ 1 billion in appropriations in 2022, and $ 1.5 billion in 2023, and as of June 2023 it

3276-621: The Wuhan Institute of Virology (WIV)—which has been at the center of speculation since early 2020 that SARS-CoV-2 may have escaped in a lab incident. Under political pressure, the NIH withdrew funding to EcoHealth Alliance in July 2020. On February 13, 2012, the National Institutes of Health (NIH) announced a new group of individuals assigned to research pain. This committee is composed of researchers from different organizations and will focus to "coordinate pain research activities across

3367-414: The cytosol of a cell. There are two types of metabolic pathways that are characterized by their ability to either synthesize molecules with the utilization of energy ( anabolic pathway ), or break down complex molecules and release energy in the process ( catabolic pathway ). The two pathways complement each other in that the energy released from one is used up by the other. The degradative process of

3458-484: The lipid bilayer . The regulation methods are based on experiments involving 13C-labeling , which is then analyzed by nuclear magnetic resonance (NMR) or gas chromatography–mass spectrometry (GC–MS) –derived mass compositions. The aforementioned techniques synthesize a statistical interpretation of mass distribution in proteinogenic amino acids to the catalytic activities of enzymes in a cell. Metabolic pathways can be targeted for clinically therapeutic uses. Within

In 2021, the NIH contributed $ 4,395,399 towards the Accelerating COVID-19 Therapeutic Interventions and Vaccines (ACTIV) program. Following the outbreak of the COVID-19 pandemic , NIH-funded EcoHealth Alliance has been the subject of controversy and increased scrutiny due to its ties to

3640-521: The product of one enzyme acts as the substrate for the next. However, side products are considered waste and removed from the cell. Different metabolic pathways function in the position within a eukaryotic cell and the significance of the pathway in the given compartment of the cell. For instance, the electron transport chain and oxidative phosphorylation all take place in the mitochondrial membrane . In contrast, glycolysis , pentose phosphate pathway , and fatty acid biosynthesis all occur in

3731-601: The 1960s, virologist and cancer researcher Chester M. Southam injected HeLa cancer cells into patients at the Jewish Chronic Disease Hospital. When three doctors resigned after refusing to inject patients without their consent, the experiment gained considerable media attention. The NIH was a major source of funding for Southam's research and had required all research involving human subjects to obtain their consent prior to any experimentation. Upon investigating all of their grantee institutions,

4004-772: The Director. NIH submits its budget request to the Department of Health and Human Services (HHS), and the HHS considers this request as a portion of its budget. Many adjustments and appeals occur between NIH and HHS before the agency submits NIH's budget request to the Office of Management and Budget (OMB). OMB determines what amounts and research areas are approved for incorporation into the President's final budget. The President then sends NIH's budget request to Congress in February for

4095-482: The IRP had 1,200 principal investigators and more than 4,000 postdoctoral fellows in basic, translational, and clinical research, being the largest biomedical research institution in the world, while, as of 2003, the extramural arm provided 28% of biomedical research funding spent annually in the U.S., or about US$ 26.4 billion. The NIH comprises 27 separate institutes and centers of different biomedical disciplines and

4186-560: The NIH Toolbox was rolled out to the research community. NIH Toolbox assessments are based, where possible, on Item Response Theory and adapted for testing by computer. NIH sponsors the Database of Genotypes and Phenotypes (dbGaP), a repository of information produced by studies investigating the interaction of genotype and phenotype. The information includes phenotypes, molecular assay data, analyses and documents. Summary-level data

4277-461: The NIH budget doubled. For a time, funding essentially remained flat, and for seven years following the financial crisis, the NIH budget struggled to keep up with inflation. In 1999 Congress increased the NIH's budget by $ 2.3 billion to $ 17.2 billion in 2000. In 2009 Congress again increased the NIH budget to $ 31 billion in 2010. In 2017 and 2018, Congress passed laws with bipartisan support that substantially increasing appropriations for NIH, which

4368-437: The NIH but may apply for funding. Throughout the history of the NIH, the amount of funding received has increased, but the proportion to each IC remains relatively constant. The individual ICs then decide who will receive the grant money and how much will be allotted. Policy changes on who receives funding significantly affect researchers. For example, the NIH has recently attempted to approve more first-time NIH R01 applicants or

4459-420: The NIH discovered that the majority of them did not protect the rights of human subjects. From then on, the NIH has required all grantee institutions to approve any research proposals involving human experimentation with review boards. In 1967, the Division of Regional Medical Programs was created to administer grants for research for heart disease, cancer, and strokes . That same year, the NIH director lobbied

4550-429: The NIH has often been a source of contention in Congress, serving as a proxy for the political currents of the time. In 1992, the NIH encompassed nearly 1 percent of the federal government's operating budget and controlled more than 50 percent of all funding for health research, and 85 percent of all funding for health studies in universities. While government funding for research in other disciplines has been increasing at

4641-670: The NIH intramural research program has been a source of many pivotal scientific and medical discoveries. Some of these include: In September 2006, the NIH Blueprint for Neuroscience Research started a contract for the NIH Toolbox for the Assessment of Neurological and Behavioral Function to develop a set of state-of-the-art measurement tools to enhance collection of data in large cohort studies. Additionally, most organisms can perform more efficient aerobic respiration through the citric acid cycle and oxidative phosphorylation . Additionally plants , algae and cyanobacteria are able to use sunlight to anabolically synthesize compounds from non-living matter by photosynthesis . In contrast to catabolic pathways, anabolic pathways require an energy input to construct macromolecules such as polypeptides, nucleic acids, proteins, polysaccharides, and lipids. The isolated reaction of anabolism

Similarly, the 13,000 to 14,000 R21 applications had a funding success of 13-14% during the same period. In FY 2016, the total number of grant applications received by the NIH was 54,220, with approximately 19% being awarded funding. Institutes have varying funding rates. The National Cancer Institute awarded funding to 12% of applicants, while

6461-1325: The movement of electrons through the ETC. The substrate-level phosphorylation that occurs at ATP synthase can also be directly inhibited, preventing the formation of ATP that is necessary to supply energy for cancer cell proliferation. Some of these inhibitors, such as lonidamine and atovaquone , which inhibit Complex II and Complex III, respectively, are currently undergoing clinical trials for FDA approval. Other non-FDA-approved inhibitors have still shown experimental success in vitro. Heme , an important prosthetic group present in Complexes I, II, and IV can also be targeted, since heme biosynthesis and uptake have been correlated with increased cancer progression. Various molecules can inhibit heme via different mechanisms. For instance, succinylacetone has been shown to decrease heme concentrations by inhibiting δ-aminolevulinic acid in murine erythroleukemia cells. The primary structure of heme-sequestering peptides, such as HSP1 and HSP2, can be modified to downregulate heme concentrations and reduce proliferation of non-small lung cancer cells. The tricarboxylic acid cycle (TCA) and glutaminolysis can also be targeted for cancer treatment, since they are essential for

6552-466: The next fiscal year's allocations. The House and Senate Appropriations Subcommittees deliberate and by fall, Congress usually appropriates funding. This process takes approximately 18 months before the NIH can allocate any actual funds. When a government shutdown occurs, the NIH continues to treat people who are already enrolled in clinical trials , but does not start any new clinical trials and does not admit new patients who are not already enrolled in

6643-539: The official date of publication. The NIH Public Access Policy was the first public access mandate for a U.S. public funding agency. In 2000, the Joint Economic Committee of Congress reported NIH research, which was funded at $ 16 billion a year in 2000, that some econometric studies had given a rate of return of 25 to 40 percent per year by reducing the economic cost of illness in the US. It found that of

6734-589: The portal display data for phenotypes, biological processes and pathways related to the disease category. Pages are also supplied to give users access to information about rat strains used as models for one or more diseases in the category, tools that could be used to analyze the data and additional resources related to the disease category. RGD's Pathway resources include a Pathway Ontology of pathway terms (developed and maintained at RGD, encompassing not only metabolic pathways but also disease, drug, regulatory and signaling pathways), as well as interactive diagrams of

6825-411: The portal display data for phenotypes, biological processes and pathways related to the disease category. Pages are also supplied to give users access to information about rat strains used as models for one or more diseases in the category, tools that could be used to analyze the data and additional resources related to the disease category. Further, access to the RGD's Multi-Ontology Enrichment Tool (MOET)

6916-654: The programs and activities of all NIH components. The NIH Director plays an active role in shaping the agency's activities and outlook. The Director is responsible for providing leadership to the Institutes and Centers by identifying needs and opportunities, especially in efforts involving multiple Institutes. Within the Director's Office is the Division of Program Coordination, Planning and Strategic Initiatives with 12 divisions including: The Agency Intramural Research Integrity Officer "is directly responsible for overseeing

7007-684: The rat strains produced in these studies via pages about the PhysGen Knockout project and the MCW Gene Editing Rat Resource Center (GERRC) , accessed from RGD page headers. Funding for both the PhysGenKO project and the GERRC came from the National Heart Lung and Blood Institute (NHLBI). The stated goal of both projects is to produce rats with alterations in one or more specific genes related to

7098-468: The research grant applications of young scientists. To encourage the participation of young scientists, the application process has been shortened and made easier. In addition, first-time applicants are being offered more funding for their research grants than those who have received grants in the past. In 2011 and 2012, the Department of Health and Human Services Office of Inspector General published

7189-517: The resolution of all research misconduct allegations involving intramural research , and for promoting research integrity within the NIH Office of Intramural Research (OIR)." There is a Division of Extramural Activities, which has its own Director. The Office of Ethics has its own Director, as does the Office of Global Research. Intramural research is primarily conducted at the main campus in Bethesda, Maryland , and Rockville, Maryland , and

7280-483: The right is an illustration of the amphibolic properties of the TCA cycle. The glyoxylate shunt pathway is an alternative to the tricarboxylic acid (TCA) cycle , for it redirects the pathway of TCA to prevent full oxidation of carbon compounds, and to preserve high energy carbon sources as future energy sources. This pathway occurs only in plants and bacteria and transpires in the absence of glucose molecules. The flux of

7371-425: The same name. The six major data types in RGD are as follows: RGD's Genome tools include both software tools developed at RGD and tools from third party sources. RGD develops web-based tools designed to use the data stored in the RGD database for analyses in rat and across species. These include: RGD offers several third party software tools that have been adapted for use on the website utilizing data stored in

7462-464: The scientific community. A secondary, but critical goal is to provide curation of mapped positions for quantitative trait loci, known mutations and other phenotypic data. The rat continues to be extensively used by researchers as a model organism for investigating pharmacology, toxicology, general physiology and the biology and pathophysiology of disease. In recent years, there has been a rapid increase in rat genetic and genomic data. In addition to this,

7553-695: The surrounding communities. The Bayview Campus in Baltimore , Maryland houses the research programs of the National Institute on Aging , National Institute on Drug Abuse , and National Human Genome Research Institute with nearly 1,000 scientists and support staff. The Frederick National Laboratory in Frederick, MD and the nearby Riverside Research Park, houses many components of the National Cancer Institute , including

7644-411: The survival and proliferation of cancer cells. Ivosidenib and enasidenib , two FDA-approved cancer treatments, can arrest the TCA cycle of cancer cells by inhibiting isocitrate dehydrogenase-1 (IDH1) and isocitrate dehydrogenase-2 (IDH2), respectively. Ivosidenib is specific to acute myeloid leukemia (AML) and cholangiocarcinoma, whereas enasidenib is specific to just acute myeloid leukemia (AML). In

7735-533: The taxpayers, making them the primary beneficiaries of advances in research. Thus, the general public is a key stakeholder in the decisions resulting from the NIH funding policy. However, some in the general public do not feel their interests are being represented, and individuals have formed patient advocacy groups to represent their own interests. Important stakeholders of the NIH funding policy include researchers and scientists. Extramural researchers differ from intramural researchers in that they are not employed by

7826-733: The top floor of the Butler Building in Washington, D.C. In 1904, it moved again to a new campus at the Old Naval Observatory , which grew to include five major buildings. In 1901, the Division of Scientific Research was formed, which included the Hygienic Laboratory as well as other research offices of the Marine Hospital Service. In 1912, the Marine Hospital Service became the Public Health Service (PHS). In 1922, PHS established

7917-490: The whole animal or systems level. Until recently, direct, specific genomic manipulations in the rat were not possible. However, with the rise of technologies such as Zinc finger nuclease - and CRISPR -based mutagenesis techniques, that is no longer the case. Groups producing rat gene knockouts and other types of genetically modified rats include the Human and Molecular Genetics Center at MCW. RGD links to information about

8008-670: Was 37.3 billion dollars annually in FY2018. Researchers at universities or other institutions outside of NIH can apply for research project grants (RPGs) from the NIH. There are numerous funding mechanisms for different project types (e.g., basic research, clinical research, etc.) and career stages (e.g., early career, postdoc fellowships, etc.). The NIH regularly issues "requests for applications" (RFAs), e.g., on specific programmatic priorities or timely medical problems (such as Zika virus research in early 2016). In addition, researchers can apply for "investigator-initiated grants" whose subject

8099-455: Was appointed as scientific member of the group. To allocate funds, the NIH must first obtain its budget from Congress. This process begins with institute and center (IC) leaders collaborating with scientists to determine the most important and promising research areas within their fields. IC leaders discuss research areas with NIH management who then develops a budget request for continuing projects, new research proposals, and new initiatives from

8190-561: Was formerly part. In 1938, NIH moved to its current campus in Bethesda, Maryland . Over the next few decades, Congress would markedly increase funding of the NIH, and various institutes and centers within the NIH were created for specific research programs. In 1944, the Public Health Service Act was approved, and the National Cancer Institute became a division of NIH. In 1948, the name changed from National Institute of Health to National Institutes of Health. In

8281-576: Was reported in newspaper articles from 1981, no funding was provided for research on the disease. In 1984 National Cancer Institute scientists found implications that "variants of a human cancer virus called HTLV-III are the primary cause of acquired immunodeficiency syndrome (AIDS)," a new epidemic that gripped the nation. In 1992, the NIH encompassed nearly 1 percent of the federal government's operating budget and controlled more than 50 percent of all funding for health research and 85 percent of all funding for health studies in universities. From 1993 to 2001
