Bacteriological water analysis is a method of analysing water to estimate the numbers of bacteria present and, if needed, to find out what sort of bacteria they are. It represents one aspect of water quality . It is a microbiological analytical procedure which uses samples of water and from these samples determines the concentration of bacteria. It is then possible to draw inferences about the suitability of the water for use from these concentrations. This process is used, for example, to routinely confirm that water is safe for human consumption or that bathing and recreational waters are safe to use.

63-536: Dharan ( Nepali : धरान ) is a sub-metropolitan city in Sunsari District of Koshi Province , in eastern Nepal, which was established as a fourth municipality in the Kingdom in 1958. It is the third most populous city in eastern Nepal after Biratnagar and Itahari . The Nepali word "dharan" means a saw pit. The rainforest from which the tree trunks came is still just on the edge of the city. Much later

126-407: A government institution established in 1913 ( B.S. 1970) for advancement of Gorkha Bhasa, renamed itself as Nepali Bhasa Prakashini Samiti (Nepali Language Publishing Committee) in 1933 (B.S. 1990), which is currently known as Sajha Prakashan . Conversely, the term Gorkhali in the former national anthem entitled " Shriman Gambhir " was changed to Nepali in 1951. However, the term Nepali

189-657: A bacteriologist for the Royal Commission on Sewage Disposal in the United Kingdom . Endo agar contains peptone, lactose, dipotassium phosphate , agar, sodium sulfite, basic fuchsin and was originally developed for the isolation of Salmonella typhi , but is now commonly used in water analysis. As in MacConkey agar, coliform organisms ferment the lactose, and the colonies become red. Non-lactose-fermenting organisms produce clear, colourless colonies against

252-471: A close connect, subsequently, the language became the lingua franca in the area. As a result, the grammar became simplified, vocabulary was expanded, and its phonology was softened, after it was syncretised, Nepali lost much of the complex declensional system present in the older languages. In the Kathmandu Valley (then known as Nepal Mandala ), Nepali language inscriptions can be seen during

315-439: A direct measure of biological concentration and health. ATP is quantified by measuring the light produced through its reaction with the naturally occurring enzyme firefly luciferase using a luminometer . The amount of light produced is directly proportional to the amount of biological energy present in the sample. Second generation ATP tests are specifically designed for water, wastewater and industrial applications where, for

378-506: A large part of the Eastern hills left the plains with the ascent of Sanghuri Danra. In the days when villagers in the hills made their annual trek to sell and buy goods this was a natural location for a market where hills and Terai met. In 1952 the construction of the Kosi barrage began and a narrow gauge railway was built to take stone from Phusre loaded at a locality now known as "Railway" to

441-460: A rectangular hollow dug under the ground and a low platform upon it for sawing logs with a big saw; one person on the platform and other below it. And, after the establishment of Dharan city, logs used to be widely split up here in "Dharan". It took its present shape when Bijaypur village panchayat and Ghopa village panchayat were merged into Dharan municipality in 2035 BS. language in Dharan At

504-649: A short period of a hundred years in the 19th century. This literary explosion was fuelled by Adhyatma Ramayana ; Sundarananda Bara (1833); Birsikka, an anonymous collection of folk tales; and a version of the ancient Indian epic Ramayana by Bhanubhakta Acharya (d. 1868). The contribution of trio-laureates Lekhnath Paudyal , Laxmi Prasad Devkota , and Balkrishna Sama took Nepali to the level of other world languages. The contribution of expatriate writers outside Nepal, especially in Darjeeling and Varanasi in India,

567-482: A sloping agar surface. Colonies that develop in the body of the medium can be counted by eye after incubation. The total number of colonies is referred to as the total viable count (TVC). The unit of measurement is cfu/ml (or colony forming units per millilitre) and relates to the original sample. Calculation of this is a multiple of the counted number of colonies multiplied by the dilution used. When samples show elevated levels of indicator bacteria, further analysis

630-593: A standardised prose in the Lal mohar (royal charter)—documents related to the Nepalese Kingdom dealing with diplomatic writings, tax, and administrative records. The language of the Lal mohar is nearly modern with some minor differences in grammar and with a pre-modern orthography. Few changes including changing Kari (करि) to Gari (गरि) and merging Hunu (हुनु) with cha (छ) to create huncha (हुन्छ) were done. The most prominent work written during this time

693-435: A tiny inverted tube called a Durham tube in each sample tube. The Durham inverted tube catches any gas produced. The production of gas at 37 degrees Celsius is a strong indication of the presence of Escherichia coli . An ATP test is the process of rapidly measuring active microorganisms in water through detection adenosine triphosphate (ATP). ATP is a molecule found only in and around living cells, and as such it gives

756-405: Is also a separate highest level honorific, which was used to refer to members of the royal family, and by the royals among themselves. Like all modern Indo-Aryan languages, Nepali grammar has syncretised heavily, losing much of the complex declensional system present in the older languages. Instead, it relies heavily on periphrasis , a marginal verbal feature of older Indo-Aryan languages. Nepali

819-671: Is also notable. Nepali-language speakers are rapidly migrating around the globe in last a couple of decades and many books of Nepali language literature are published from different corners of the world. Diasporic literature has developed new ways of thinking and created a new branch in Nepali language literature. Dialects of Nepali include Acchami, Baitadeli, Bajhangi, Bajurali, Bheri, Dadeldhuri, Dailekhi, Darchulali, Darchuli, Gandakeli, Humli, Purbeli, and Soradi. These dialects can be distinct from Standard Nepali. Mutual intelligibility between Baitadeli, Bajhangi, Bajurali (Bajura), Humli and Acchami

882-630: Is an Indo-Aryan language native to the Himalayas region of South Asia . It is the official, and most widely spoken, language of Nepal , where it also serves as a lingua franca . Nepali has official status in the Indian state of Sikkim and in the Gorkhaland Territorial Administration of West Bengal . It is spoken by about a quarter of Bhutan 's population. Nepali also has a significant number of speakers in

945-627: Is believed to have originated around 500 years ago with the mass migration of a branch of Khas people from the Karnali - Bheri - Seti eastward to settle in lower valleys of the Karnali and the Gandaki basin. During the times of Sena dynasty , who ruled a vast area in Terai and central hills of Nepal, Nepali language became influenced by the Indian languages including Awadhi , Bhojpuri , Braj Bhasha and Maithili . Nepali speakers and Senas had

1008-773: Is generally written in Devanagari script. In certain regions, the Tibetan script was also used in regions with predominantly Tibetic population, with common Tibetan expressions and pronunciation. In the section below Nepali is represented in Latin transliteration using the IAST scheme and IPA . The chief features are: subscript dots for retroflex consonants ; macrons for etymologically, contrastively long vowels ; h denoting aspirated plosives . Tildes denote nasalised vowels . Nepali developed significant literature within

1071-418: Is incubated at 22 °C and for 24 hours and a second set at 37 °C for 24 hours. The composition of the nutrient usually includes reagents that resist the growth of non-target organisms and make the target organism easily identified, often by a colour change in the medium. Some recent methods include a fluorescent agent so that counting of the colonies can be automated. At the end of the incubation period

1134-613: Is low. The dialect of the Nepali language spoken in Karnali Province is not mutually intelligible with Standard Nepali. The language is known by its old name as Khas Bhasa in Karnali. The following is a sample text in Nepali, of Article 1 of the Universal Declaration of Human Rights , with a transliteration (IAST) and transcription ( IPA ). 1197-824: Is often in free variation with [õ]. Nepali has ten diphthongs : /ui̯/, /iu̯/, /ei̯/, /eu̯/, /oi̯/, /ou̯/, /ʌi̯/, /ʌu̯/, /ai̯/, and /au̯/. [j] and [w] are nonsyllabic allophones of [i] and [u], respectively. Every consonant except [j], [w], and /ɦ/ has a geminate counterpart between vowels. /ɳ/ and /ʃ/ also exist in some loanwords such as /baɳ/ बाण "arrow" and /nareʃ/ नरेश "king", but these sounds are sometimes replaced with native Nepali phonemes. The murmured stops may lose their breathy-voice between vowels and word-finally. Non-geminate aspirated and murmured stops may also become fricatives, with /pʰ/ as [ ɸ ], /bʱ/ as [ β ], /kʰ/ as [ x ], and /ɡʱ/ as [ ɣ ]. Examples of this are /sʌpʰa/ 'clean' becoming [sʌɸa] and /ʌɡʱaɖi/ 'before' becoming [ʌɣaɽi]. Typically, sounds transcribed with

1260-839: Is often undertaken to look for specific pathogenic bacteria. Species commonly investigated in the temperate zone include Salmonella typhi and Salmonella Typhimurium . Depending on the likely source of contamination investigation may also extend to organisms such as Cryptosporidium spp . In tropical areas analysis of Vibrio cholerae is also routinely undertaken. MacConkey agar is culture medium designed to grow Gram-negative bacteria and stain them for lactose fermentation. It contains bile salts (to inhibit most Gram-positive bacteria), crystal violet dye (which also inhibits certain Gram-positive bacteria), neutral red dye (which stains microbes fermenting lactose), lactose and peptone . Alfred Theodore MacConkey developed it while working as

1323-459: Is otherwise similar to conventional total plate counts. Membranes have a printed millimetre grid printed on and can be reliably used to count the number of colonies under a binocular microscope. When the analysis is looking for bacterial species that grow poorly in air, the initial analysis is done by mixing serial dilutions of the sample in liquid nutrient agar which is then poured into bottles which are then sealed and laid on their sides to produce

1512-538: Is the second most commonly spoken language other than English. Allawah and Hurstville have third most Nepali speaking population in New South Wales . There are regular Nepali language News papers and Magazines in Australia. Vowels and consonants are outlined in the tables below. Nepali distinguishes six oral vowels and five nasal vowels . /o/ does not have a phonemic nasal counterpart, although it

1575-775: Is therefore reasonable to surmise that if indicator organism levels are low, then pathogen levels will be very much lower or absent. Judgements as to suitability of water for use are based on very extensive precedents and relate to the probability of any sample population of bacteria being able to be infective at a reasonable statistical level of confidence. Analysis is usually performed using culture , biochemical and sometimes optical methods. When indicator organisms levels exceed pre-set triggers, specific analysis for pathogens may then be undertaken and these can be quickly detected (where suspected) using specific culture methods or molecular biology . The most reliable methods are direct plate count method and membrane filtration method. mEndo Agar

1638-546: Is used in the membrane filtration while VRBA Agar is used in the direct plate count method. VRBA stands for violet red bile agar. A media that contains bile salts which promotes the growth of gram negative and has inhibitory characteristic to gram positive although not complete inhibitory. These media contain lactose which is usually fermented by lactose fermenting bacteria producing colonies that can be identified and characterised. Lactose fermenting produce colored colonies while non lactose fermenting produce colorless ones. Because

1701-452: Is used where no respect is due, medium honorific is used to signify equal status or neutrality, and high honorific signifies respect. Like all modern Indo-Aryan languages, Nepali grammar has syncretised heavily, losing much of the complex declensional system present in the older languages. Nepali was originally spoken by the Khas people , an Indo-Aryan ethno-linguistic group native to

2394-402: The action trigger levels for different waters vary depending on the use made of the water. Whilst very stringent levels apply to drinking water , more relaxed levels apply to marine bathing waters, where much lower volumes of water are expected to be ingested by users. The common feature of all these routine screening procedures is that the primary analysis is for indicator organisms rather than

2457-416: The analysis is always based on a very small sample taken from a very large volume of water, all methods rely on statistical principles. One of the oldest methods is called the multiple tube method. In this method a measured sub-sample (perhaps 10 ml) is diluted with 100 ml of sterile growth medium and an aliquot of 10 ml is then decanted into each of ten tubes. The remaining 10 ml is then diluted again and
